The Port Authority Bus Terminal is the destination and launching point for most buses in and out of New York - with the exception of the Chinatown buses, which naturally leave from Chinatown. It is located at 42nd Street and 8th Avenue, a stone's throw from Times Square (alighting from the 1/2/3 subway at 42nd St./Times Sq. station is the easiest way to get there from Columbia). Port Authority, as it's usually called, is renown for its seediness, although it's surprisingly decent inside - no worse, at least, than Penn Station a bit further downtown.
